Ipamorelin is a synthetic pentapeptide first described in the 1990s by researchers at Novo Nordisk during a program to develop selective growth hormone secretagogues. Its sequence is Aib-His-D-2-Nal-D-Phe-Lys-NH2, incorporating two non-natural residues, alpha-aminoisobutyric acid and D-2-naphthylalanine. The C-terminus is amidated, and the material is supplied as a white lyophilized powder. The molecular formula is C38H49N9O5 and the monoisotopic mass is approximately 711.85 daltons. The short chain and modified residues give it greater resistance to enzymatic degradation than many larger peptide hormones.
At the molecular level, ipamorelin acts as an agonist at the growth hormone secretagogue receptor type 1a, the same G protein-coupled receptor that binds ghrelin. Receptor activation couples to Gq/11 signaling, raising intracellular calcium through inositol trisphosphate and diacylglycerol, which in turn promotes exocytosis of growth hormone from pituitary somatotroph cells. Ipamorelin binds this receptor with high affinity and shows weak activity at other secretagogue-related targets in vitro. Its action requires the intact receptor and is not reversed by growth hormone-releasing hormone antagonists.

Signal transduction begins when the peptide binds GHSR-1a on pituitary somatotrophs. The receptor couples to Gq/11 proteins, activating phospholipase C, which cleaves phosphatidylinositol bisphosphate into inositol trisphosphate and diacylglycerol. Inositol trisphosphate releases calcium from intracellular stores, and the resulting rise in cytosolic calcium drives growth hormone vesicle fusion. Concurrent Gs coupling and cyclic AMP elevation have also been reported, and the relative contribution of each arm to the overall secretory response is not fully settled.
Structural features distinguish the molecule from earlier secretagogues. An alpha-aminoisobutyric acid residue near the N-terminus and a D-naphthylalanine substitution increase receptor affinity, while C-terminal amidation improves resistance to exopeptidases. These modifications are associated with reduced stimulation of appetite and of the hypothalamic-pituitary-adrenal axis compared with hexarelin or growth hormone releasing peptide-6. Whether the same profile applies at every dose level studied is a matter of ongoing investigation rather than settled consensus.
Lyophilized material is generally held at minus twenty degrees Celsius or lower, protected from moisture and light. Repeated excursions to room temperature cause condensation inside the vial and gradual moisture uptake, both of which shorten shelf life. Containers should be allowed to equilibrate before opening so that water does not condense on the solid. Dividing a batch into single-use aliquots reduces freeze-thaw cycling. Solid peptide handled this way is usually considered stable for months to years, while the same material in solution degrades on a much shorter timescale.
Common solvents for laboratory work include water, buffered saline, and dimethyl sulfoxide. Once dissolved, the peptide is exposed to hydrolysis and oxidation, and alkaline conditions accelerate breakdown. Low-binding plasticware and the addition of a carrier protein reduce losses to container surfaces, which can otherwise be substantial at low concentrations. Solutions are typically kept cold and used within days. Investigators working with the compound generally prepare fresh working dilutions rather than storing dilute stocks, and they avoid repeated warming of the same vial.
Reversed-phase high-performance liquid chromatography is the standard method for purity assessment, most often on a C18 column with a water and acetonitrile gradient and trifluoroacetic acid or formic acid as an ion-pairing agent. Mass spectrometry by electrospray or matrix-assisted laser desorption confirms the expected mass and reveals truncated or modified sequences. Amino acid analysis and sequencing provide orthogonal structural evidence. Typical impurities include deletion sequences, oxidized products, and dimeric species. Detection wavelength, usually 214 or 220 nanometers, should be reported because response factors differ between peptides.
=== Metallocenophanes === Metallocenophanes feature linking of the cyclopentadienyl or polyarenyl rings by the introduction of one or more heteroannular bridges. Some of these compounds undergo thermal ring-opening polymerizations to give soluble high molecular weight polymers with transition metals in the polymer backbone. Ansa-metallocenes are derivatives of metallocenes with an intramolecular bridge between the two cyclopentadienyl rings.
In March 2020, UPS expanded its autonomous trips with TuSimple by adding an extra route between Phoenix and El Paso, Texas. In January 2021, UPS announced it had agreed to sell UPS Freight, its less-than-truckload freight business, to TFI International, a Canadian transport and logistics company, for $800 million. UPS said the move would allow it to focus on small-package delivery. At the time of the sale, UPS Freight had about 14,500 employees, approximately 11,000 of them represented by the Teamsters union, and generated an estimated $3.15 billion in revenue in 2020 offering services across the US, Canada, and Mexico. TFI had reported $4.1 billion in revenue in 2019 and already operated truckload and LTL services in Canada. The acquisition was completed in April and UPS Freight was renamed TForce Freight. In 2021, following the company's shift to target smaller customers to boost profits during the COVID-19 pandemic, UPS reported a 21% jump in their fourth quarter sales to $24.9 billion. CEO Carol Tomé reported that Amazon paid UPS $11.3 billion in shipping in 2020, accounting for 13.3% of the company's revenue. In September 2021, UPS entered into an agreement to acquire Roadie for an undisclosed amount with the transaction expected to be closed in the fourth quarter. In May 2022, UPS acquired logistics company Delivery Solutions, a key partner for Walmart's GoLocal network. In November 2022, it was announced UPS had acquired the healthcare focused, Europe-based warehousing and temperature-controlled transport company, Bomi Group.
Since its 1947 independence, India has significantly reduced historically endemic diseases, though some remain intractable. The country was certified smallpox-free in April 1977 following two years of surveillance after its last Variola major case. It was officially certified polio-free by the World Health Organisation on 27 March 2014, after successfully completing three consecutive years without recorded cases of wild poliovirus. In contrast, cholera remains endemic to Ganges-Meghna Delta estuaries, resulting in seasonal outbreaks caused by monsoons and water supply network shortcomings. Tuberculosis is a major concern; India bears a quarter of the global tuberculosis disease burden. Historical malaria cases have dropped from millions to hundreds of thousands annually, but India experiences localised seasonal increases caused by drug-resistant variants of the Plasmodium falciparum and Plasmodium vivax vectors. Patients in India often face a stark choice: use basic, free, publicly funded health care or pay high out-of-pocket fees for better-equipped care from private doctors and hospitals. This divide is particularly severe outside cities, as public clinics in rural areas rarely have enough doctors or nurses to meet local demands. When serious medical emergencies occur, poorer families are often forced to sell land or take out high-interest loans to pay for private care. This resulting medical debt pushes approximately 55 million people into poverty every year.

Nasopharyngeal angiofibromas, also termed juvenile nasopharyngeal angiofibromas, fibromatous hamartomas, or angiofibromatous hamartoma of the nasal cavity, are large benign tumors (average size 5.9 cm in one study) that develop almost exclusively in males aged 9 to 36 years old. They commonly arise in the nasopharynx (i.e. upper part of the throat that lies behind the nose) and typically have attachments to the sphenopalatine foramen, clivus, and/or root of the pterygoid processes of the sphenoid bone. These tumors may expand into various other nearby structures including the cranial cavity. Nasopharyngeal angiofibromas are highly vascularized tumors consisting of fibroblasts (i.e. connective tissue cells) in a dense collagen matrix (i.e. tissue background). Studies have suggested that these tumors are due to the expression of male sex hormones (i.e. androgens and progesterones), genetic factors, molecular alterations (i.e. changes in the normal characteristics of cells that lead to abnormal cell growth), and/or human papillomavirus infection.
=== Bethlem myopathy 2 === (Collagen XII gene) See Bethlem myopathy 2 Clinical synopsis on OMIM: 616471 In Bethlem myopathy 2, there is phenotypic variability. In one family, the only notable finding on T1-weighted MR images (used to detect fatty infiltration) was atrophy of the rectus femoris muscles of the thigh, with the degree of atrophy matching the severity of the disease, but no fatty infiltration. In another family, only the more severely affected older patient showed significant abnormality, by having symmetrical fatty atrophy of the femoral quadriceps of the thigh, the adductor and medial gastrocnemius muscles of the calf; as well as asymmetrical fatty atrophy of the adductor longus of the thigh. No muscle hypertrophy was reported and the muscles of the patients without fatty atrophy appeared normal. Bethlem myopathy 2 also differs by including the possibility of scapula winging, pectus excavatum, stooped posture, kyphosis (hunchback), micrognathia, retrognathia, and a high-arched palate. Childhood muscle weakness improves in teen years, but muscle weakness returns by the third decade of life.
== Epidemiology == Ehlers–Danlos syndromes are estimated to occur in about one in 5,000 births worldwide. Initially, prevalence estimates ranged from one in 250,000 to 500,000 people, but these were soon found to be low, as medical professionals became more adept at diagnosis. EDS may be far more common than the currently accepted estimate due to the wide range of severities with which the disorder presents. The prevalence of the disorders differs dramatically. The most common is hypermobile EDS, followed by classical EDS. The others are very rare. For example, fewer than 10 infants and children with dermatosparaxis EDS have been described worldwide. Some types of EDS are more common in Ashkenazi Jews. For example, the chance of being a carrier for dermatosparaxis EDS is one in 2,000 in the general population but one in 248 among Ashkenazi Jews. Some recent studies have found that EDS is significantly more prevalent in people who identify as transgender or gender-diverse, or who suffer from gender dysphoria. One review, of a pediatric EDS clinic in the American Midwest between 2020 and 2022, found that 17% of patients identified as trans or gender-diverse, 89% of whom were assigned female at birth. By comparison, roughly 1–2% of adolescents identify as trans or gender-diverse in the US overall. In a 2020 study, among adults undergoing gender-affirming surgery, 2.6% had a diagnosis of EDS—130 times the highest reported prevalence of EDS in the general population.
== Part 2 == The second part runs approximately two-and-a-half hours and follows Metallica at the start of their Wherever We May Roam Tour in Europe through to the Guns N' Roses/Metallica Stadium Tour of 1992. Included are performances of "For Whom the Bell Tolls" from Donington on August 17, 1991, "Enter Sandman" from the MTV Video Music Awards on September 5, "Harvester of Sorrow" from Moscow on September 28, "Sad but True" from the Day on the Green festival in Oakland, California on October 12, "Enter Sandman" from the Freddie Mercury Tribute Concert on April 20, 1992, and "Nothing Else Matters" from Phoenix on August 25. Jason Newsted after a concert in Portland, Maine, is seen making sandwiches to take back to a hotel with him instead of ordering room-service. His response to being called a cheapskate is "I got plans for those millions and it ain't for fucking sandwiches!" Sebastian Bach is seen joking with Slash and Lars Ulrich in Metallica's dressing room at RFK Stadium in Washington, D.C. prior to the opening concert of the joint tour with Guns N' Roses. Bach humorously imitates MTV host Riki Rachtman, and former Metallica guitarist Dave Mustaine. The band also encounter Spinal Tap, who jokingly ask them about the similarities between their album's covers. This part of the documentary also includes two music videos:

Consumption of these peaks during cultural festivities such as the Mongolian lunar new year (in spring). The time of this celebration is called the "white month", which indicates that milk products (called "white food" together with starchy vegetables, in comparison to meat products, called "black food") are a central part of this tradition. The purpose of these festivities is to "close" the past year – clean the house or the yurt, honor the animals for having provided their food, and prepare everything for the coming summer season – to be ready to "open" the new year. Consuming white food in this festive context is a way to connect to the past and to a national identity, which is the Mongol Empire personified by Genghis Khan. During the time of this empire, the fermented mare milk was the drink to honor and thank warriors and leading persons, it was not meant for everybody. Although it eventually became a drink for normal people, it has kept its honorable meaning. Like many other traditions, this one feels the influence of globalization. Other products, like industrial yogurt, coming mainly from China and western countries, have tended to replace it more and more, mainly in urban areas. However, in rural and poorer regions it is still of great importance. Although this chemical process had been used in food production for thousands of years, the mechanism of microbial lactic acid fermentation was not understood until much later. During the 19th century, several chemists discovered some fundamental concepts of organic chemistry.
Two years prior, the French had refused Ho's proposal of a similar status within the French Union, albeit with some restrictions on French power and the latter's eventual withdrawal from Vietnam. However, they were willing to deal with Bảo Đại as he represented a non-radical option who could rally behind him the non-Communist nationalist movement. In January 1950, France officially recognized the nominal "independence" of the unified State of Vietnam, led by Bảo Đại, as an associated state within the French Union. However, France still controlled all foreign policy, every defense issue and would have a French Union army stationed in the country with complete freedom of movement. Within the framework of the French Union, France also granted independence to the other nations in Indochina, the Kingdoms of Laos and Cambodia. In January 1949, the Vietnamese National Army was created to go along the formation of the new Vietnamese associated state. This was meant to bolster French numbers as their army found itself outnumbered by the People's Army of Vietnam at this point in the war. To this end, the CEFEO provided some of its officers to lead these new divisions.
Dephosphorylation is commonly done using calf-intestinal alkaline phosphatase (CIAP) which removes the phosphate group from the 5′ end of digested DNA, but note that CIAP is not easy to inactivate and can interfere with ligation without an additional step to remove the CIAP, thereby resulting in failure of ligation. CIAP should not be used in excessive amount and should only be used when necessary. Shrimp alkaline phosphatase (SAP) or Antarctic phosphatase (AP) are suitable alternative as they can be easily inactivated.

It is a synthetic pentapeptide belonging to the growth hormone secretagogue family. Its principal characterized target is the ghrelin receptor, also called GHS-R1a. The molecule contains non-natural amino acids and an amidated C-terminus.
Both act at the same G protein-coupled receptor, but ipamorelin is a short synthetic peptide with modified residues rather than the natural 28-amino-acid hormone. Reports describe weaker effects on appetite and on cortisol or prolactin release than those seen with ghrelin. Its resistance to enzymatic breakdown also differs from that of the natural ligand.
Receptor binding and downstream calcium signaling are well characterized in cell-based systems. Effects measured in whole organisms are less consistent across studies and species. The extent to which selective receptor behavior drives the observed hormonal profile is still debated.
